Does eNB communicates with another eNB using wireless medium or using wired medium ?

there is no explicit sentence states that both are connected via wired interface but with the context we can predict, enb finds and another ENB IP and then sends request. If both are communicating over the air then it requires any protocol to detect the message communication (because X2 is an application protocol), so it is not using any OTA communication as if it uses IP based execution it will be an wired base, and i have seen myself X2setup in wired base.
